Calculation of Absorbance Values

The absorbance value (Aλ) of near infrared spectra is calculated using dark, reference, and sample intensity values at each wavelength (λ) according to the formula: Aλ = log[(Sλ - Dλ) / (Rλ - Dλ)]. Absorbance is determined by first collecting dark readings with the light source off and reference readings with the light source on and a reflector in place. The preprocessed absorbance values are then corrected and used to build a mathematical model for analysis.

Calculation of Absorbance values

of the reflected NIRS spectra is calculated with the Dark and


Reference spectra using the following expression:

is the reference intensity at wavelength .



It is to be noted that while collecting the Dark readings, the Lamp source has to be turned
off. When collecting the Reference readings the Lamp source has to be turned on and a
perfect reflector (white tile or smooth Aluminum plate) has to be placed beneath the optical
setup.
After calculation of the absorbance value, the spectra is corrected for baseline, additive and
multiplicative errors. The preprocessed data is then used to build a mathematical model for
further analysis.



Note: Relation between Absorbance and % Transmittance

A = log (1/T)

or

A = 2 - log(%T)
