(C) An example of a strongly down-regulated gene near a superenhancer. In untreated cells, a series of cohesin-associated loops form between the IER5L promoter and nearby superenhancers. Upon auxin treatment, these loops are lost and IER5L expression is 2.6-fold down-regulated. (Note that, unlike this example, many of the genes that are strongly down-regulated after treatment do not exhibit visible promoter-enhancer looping in untreated cells.)
