Introduction To The Microscope: Care Parts Focusing

This document provides instructions for using a microscope, including its parts and proper care, handling, and focusing techniques. Key steps are to carry the microscope with two hands, clean only with lens paper, avoid forcing knobs, store covered, and keep objects clear of the desk and cords. It explains how to place a slide on the stage under the objective lens, plug it in, use stage clips, and focus using the coarse and fine focusing knobs starting with the lowest power setting. Making a wet mount slide involves placing a thin specimen and one drop of water on a slide, then covering at an angle to avoid bubbles.

Base
• Place the Slide on the
microscope, specimen side up
• Plug into outlet
• Use Stage Clips
• Click objective lens to the
lowest (shortest) setting.
• Look into the Eyepiece
• Use the Coarse Focus
• Follow steps to focus using low power
• Click the lens to the longest objective
• Do NOT use the Coarse Focusing Knob
– That’s the big one
• Use the Fine Focus Knob until you can see
specimen clearly
– That’s the small one
Making a “Wet Mount” slide
• 1.Gather a thin slice/piece of your
specimen. If your specimen is too thick,
then the coverslip will wobble on top of
the sample like a see-saw:

Making a “Wet Mount” slide
• 2. Place ONE drop of water directly
over the specimen.
Making a “Wet Mount” slide
• 3. Placethe coverslip at a 45 degree
angle (approximately), with one edge
touching the water drop, and let go.
Making a “Wet Mount” slide
• Dropping the cover slip directly onto the
slide will cause air bubbles to be trapped
